Protein Status Modulates the Rewarding Value of Meals according to their Protein Content in Rats

Résumé

Maintenance of normal physiological functions and survival in animals and humans require a continuous supply of amino acids to tissues. Protein status is tightly controlled and might influence feeding behavior in order to prevent protein deficiency. The present study hypothesizes that reward pathways are sensitive to protein status and are involved in these processes. Wistar rats were adapted to different Meal and Diet combinations (n=6 rats per group) constituted by a normal protein (NP), a high protein (HP), or a low-protein diet (LP) feed. Results showed that rats in a situation of generous protein supply with a HP diet, in comparison to a control NP diet, had lower daily energy intake that agree with the satiating effect of protein, and this was associated to a significant lower total NAcc neuronal activation, a significant higher NAcc expression of dopamine receptor 2 (DR2) mRNA and significant differences in NAcc expression in delta (DOR) and kappa opioid receptors (KOR). In contrast, in rats in a situation of protein restriction with a LP diet in comparison to rats fed a control NP diet, rats showed a significance preference for a protein-rich meal and this was associated to higher fasted blood ghrelin level, lower blood leptin level, higher hypothalamic NPY expression and significant higher meal protein content-related NAcc neuronal activation. These results demonstrated that reward pathways are sensitive to protein status and that when protein status is deficient the activity of the reward system is positively related to the protein content of meal in relation with an appetite for protein-containing foods. These results indicate that protein sufficiency lowers the hedonic value of protein-containing meals whereas in a situation of protein deficiency the preferences for protein could be related to higher hedonic value of protein-containing meals. This is associated to a cascade of signaling processes from the periphery to the brain involving ghrelin, leptin and hypothalamic NPY activity.
